The test uses fluorescent-labeled antibodies to visualize these interactions under a microscope. It is often used in the diagnosis of autoimmune diseases such as lupus, pemphigus, and other immune-mediated conditions.

This test is typically ordered when autoimmune disease is suspected. It helps detect antibodies that target tissue antigens, indicating immune system dysfunction. For example, in cases of skin blistering diseases or systemic autoimmune disorders, the test can localize antibody binding in affected tissues.

A positive result indicates the presence of tissue-specific IgG antibodies, suggesting an autoimmune process. The pattern and intensity of the fluorescence can help in determining the specific condition. However, results must be interpreted in conjunction with clinical findings and other diagnostic tests to confirm a diagnosis.

A tissue sample is prepared on a slide, and the patient's serum is applied. If autoantibodies are present, they will bind to antigens in the tissue. Fluorescent-labeled anti-human IgG antibodies are then added, which attach to any bound patient antibodies. The slide is examined under a fluorescence microscope for diagnostic patterns.

While highly useful, this test is dependent on the skill of the technician and the quality of the sample.
