The Anti-IDH1 (H09) test is an immunohistochemistry (IHC) assay that detects the R132H mutation in the IDH1 gene. This mutation is commonly found in certain types of gliomas and other central nervous system (CNS) tumors. It helps in diagnosing tumor types, predicting clinical outcomes, and deciding targeted therapies.
IDH1 mutations are known biomarkers in gliomas and play a role in tumor metabolism and progression. Detecting this mutation helps confirm the diagnosis and has prognostic value, as tumors with IDH1 mutations typically have better clinical outcomes. Moreover, it aids in identifying patients eligible for specific IDH1-targeted therapies.
The test is recommended when a brain tumor or glioma is suspected and tissue biopsy is available. It is especially indicated during the histopathological evaluation of gliomas or astrocytomas to classify tumor subtype and grade.
A positive result confirms the presence of the R132H IDH1 mutation, supporting a diagnosis of IDH-mutant glioma. This finding can influence grading and treatment strategy. A negative result suggests the absence of the specific mutation but does not exclude other IDH mutations or gliomas.
The test is done on tumor tissue sections using the H09 antibody clone, which specifically binds to the R132H mutant protein. Visualization is done through staining techniques and interpreted by a pathologist under a microscope.
