The Sucrose Lysis Test detects red blood cell (RBC) fragility in low ionic strength environments, used for diagnosing paroxysmal nocturnal hemoglobinuria (PNH).

Patients with hemolytic anemia, unexplained hemoglobinuria, or thrombosis should consider this test.

A blood sample is mixed with sucrose solution, and the degree of RBC hemolysis is measured.

Positive results suggest increased RBC fragility and possible PNH, requiring further confirmation with flow cytometry.

Flow cytometry analysis for CD55 and CD59 is recommended to confirm the diagnosis of PNH. Treatment may include eculizumab to reduce hemolysis and thrombosis risk.
