The HAM’s Test is used to diagnose paroxysmal nocturnal hemoglobinuria (PNH), a rare blood disorder. It detects defective red blood cells that are more susceptible to destruction. This test is important for diagnosing PNH and guiding treatment options such as eculizumab.

This test is used to diagnose paroxysmal nocturnal hemoglobinuria (PNH), a rare blood disorder. It assesses the susceptibility of red blood cells to lysis in an acidified environment.
It is performed to confirm a diagnosis of PNH in patients presenting with unexplained hemolytic anemia, blood clots, or bone marrow failure.
A blood sample is mixed with acidified serum. The degree of hemolysis is then evaluated to assess the presence of abnormal red blood cells.
Significant hemolysis indicates a positive test and supports the diagnosis of PNH. However, newer flow cytometry-based tests are now preferred for accuracy.
No specific preparations are necessary for this test.
