This test is used to diagnose paroxysmal nocturnal hemoglobinuria (PNH), a rare blood disorder. It assesses the susceptibility of red blood cells to lysis in an acidified environment.

It is performed to confirm a diagnosis of PNH in patients presenting with unexplained hemolytic anemia, blood clots, or bone marrow failure.

A blood sample is mixed with acidified serum. The degree of hemolysis is then evaluated to assess the presence of abnormal red blood cells.

Significant hemolysis indicates a positive test and supports the diagnosis of PNH. However, newer flow cytometry-based tests are now preferred for accuracy.

No specific preparations are necessary for this test.
